Effects of 5-Fluorouracil on the Articular Disk of the Temporomandibular Joint of the Rat


Abstract
Histochemical studies were undertaken in respect of the effects of 5-Fluorouracil on the articular disk of the temporomandibular joint of the young rats. Experimental animals were given an intramuscular injection of 5-Fluorouracil 5 mg per kg of body weight. The animals were sacrificed 3, 5, 10 and 18 days after 5-Fluorouracil injection. The disks were dissected from temporomandibular joint and sectioned at different levels, both sagittally and parallel to the joint surfaces. The disks were fixed in neutral formalin and Bouin¢¥s fixative. They were then embedded in paraffin. The sections were stained with hematoxylin-eosin, toluidine blue, methyl green-pyronin, PAS reaction, Orcein van Gieson stain and Masson¢¥s trichrome stain. The observations performed under ordinary light microscope were summarized as follows. 1. After 5-Fluorouracil injection, general histological structures of the temporomandibular disks of the young rats were severely changed on the 5 day group; i.e. relatively decrease in number and nuclear pyknosis of the fibroblasts, capillarization and hyperemia in the submesodelial layer, and infiltration of the fat cells in the disks. 2. In the findings of the methyl green-pyronin stain, the fibroblasts were strongly reacted in the control group, but the staining intensities were more or less weak in the groups of 3 days or 5 days after 5-Fluorouracil injection. It is considerable that 5-Fluorouracil was suppressively affective in the nuclear synthesis of the fibroblasts. 3. In the Orcein van Gieson¡¯s stain and Masson trichrome stain, the collagenous fiber bundles were densely arranged in the control group, but in the groups of 3 or 5 days after 5-Fluorouracil injection the collagenous fibers were more loosely and irregularly arranged. The fine elastic fibers which were interlaced in the collagenous fiber bundles of the control rats, were not changed in the experimental groups.
